Most of the people will fly in or out from Buenos Aires, which is wicked, as BA is the cheapes airport at the moment in Southamerica.
My route started in BA, went down on through Peninsula Valdes, Bariloche, Calafate, short crossing over to Chile for Torres del Paine and then further down to the End of the World Ushuaia, from where you can get flights back to BA for a hundred US (which is cheaper as 3 days on a bus). Spent another week in BA, crossed the river to see Uruguay, then moved on to Mendoza, Cordoba, Salta, Rosario, Paraguay and further on to Iguacu falls in Brazil.

Buenos Aires: Will not go into to much detail - most is covered in travelguides anyway and nightlife is so big and changing, that it wouldnt make sense to put it down here. 
 
Here just a few off the beaten track places:
1. The floating casino -- free drinks all night long as long as you play
2. Bar Pelvis - cheap beer, jukebox, poolbillard and very friendly girls
3. Grants - all you can eat place, so much food and such good quality
 
Places to stay:
St. Nicholas Youth Hostel - good place, chilled and central
Milhouse Hostel - big party place, lots of drinking going on
 
Peninsula Valdes: Go to Puerto Madryn, rent a car and drive to the peninsula - a whole day trip. Also quite cool is to camp out at the beach there as you can hear and see the whales all night long. It doesnt matter, if you take a cheap big boat or a small one, the whales dont give a damn and jump infront of you anyway. Pretty cool stuff.
 
Bariloche: Chocolate capital of Southamerica, little Austria, mountains, lakes, forrests, snow and skiing. Really cool place to chill and do some hiking. Also perfect place to do Paragliding.
 
El Calafate: The reason to come here is the famous Perito Moreno glacier, really amazing, but dont take a tour, do it yourself, hitch or take a bus. Bring some food and have picknick there in the late afternoon for sunset and listen to the ice breaking, when all the Warmduschers are gone.
 
From there you can also go to Chalten for further trekking (Fitz Roy Area).
 
Ushuaia: The End of the World ... get a special stamp for your passport and send postcards. Its a bit of a cold and weird place, but some people like it. 2 days were enough for me though.
 
Mendoza: Wine capital of Argentina. Malbec and Syrah, get pissed on a wine cellar tour, kayaking, trekking or just chilling on the main plaza. Pretty cool place, dont miss it. Got Argentinas hottest women there.
 
Cordoba: Student town. Go there for the weekend, got brilliant nightlife, otherwise not much to do.
 
Salta: Loads of daytrips to do from there, pretty cool landscape, go and see the seven cunts (quebrada de siete conchas), best to do that by renting a car.
 
Rosario: Pretty cool city, amazing nightlife and it s supposed to host Argentina's most gorgeous women ... although Mendoza got the better density ...
 
Puerto de Iguazu: Heaps of water, amazing falls, mindblowing. Argentina side is the better side, dont miss boca del diablo.
